Hepatitis- B surface antigen (HBsAg)
Latex agglutination method
এই test এর মাধ্যমে Hepatitis B virus এর Surface Antigen এর উপস্থিতি দেখা হয়। Hepatitis B virus এর screen test করতে এবং blood donors HBV আক্রান্ত কিনা নির্ণয় করতে প্রাথমিক ভাবে এ test করা হয়। আক্রান্ত হওয়ার 4-12 সপ্তাহের মধ্যে এই Antigen এর উপস্থিতি নির্ণয় করা যায়। এই test Hepatitis B দ্বারা আক্রান্ত কিনা তা নির্নয়ের একটি কার্যকর পরীক্ষা HBsAg test এর Negative হলে বুঝতে হবে রোগীর দেহে Hepatitis B virus এর উপস্থিতি নেই। HBsAg test Positive হলে রোগী Hepatitis B virus দ্বারা আক্রান্ত বুঝায়। কোন রোগীর সবসময় HBsAg test Positive বা স্বাভাবিকের চেয়ে বেশী হলে। রোগী Hepatitis B virus এর carrier নির্দেশ করে।
দেহে hepatitis B virus এর উপস্থিতি নির্ণয়ের জন্য HBsAg test করা হয়।
Method: Latex agglutination method / Latex slide test.
Reagent: HBsAg-র reagent এর box এ সাধারণত থাকে:
1. HBsAg latex reagent
2. HBsAg positive control
3. HBsAg negative control
4. Disposable slide
5. Disposable mixing stick
Sample: Serum or plasma (fresh)
Test procedure (পরীক্ষা পদ্ধতি):
>> প্রথমে reagent এবং serum রুম তাপমাত্রায় আনতে হবে।
>> একটি slide এর একটি circle এ one drop HBsAg-র latex reagent নিতে হবে।
>> Reagent এর পাশে one drop (50ul) serum নিতে হবে।
>> Disposable stick দ্বারা reagent এবং serum mix করতে হবে।
>> Lab rotator এ 100rpm এ 5 minute ঘুরাতে হবে।
>> পর্যাপ্ত আলোতে agglutination হয়েছে কিনা দেখতে হবে।
>> Agglutination হলে HBsAg: Positive.
>> Agglutination না হলে HBsAg: Negative.
HBsAg reagent এর positive / negative control ব্যবহারের নিয়ম:
>> Slide এর একটি circle এ one drop HBsAg latex reagent এর সাথে one drop positive control mix করে ৫ মিনিট lab rotator এ 100 rpm এ ঘুরালে agglutination হবে অর্থাৎ positive result পাওয়া যাবে।
>> আবার one drop HBsAg latex reagent এর সাথে one drop negative control মিশিয়ে ৫ মিনিট lab rotator এ 100 rpm এ ঘুরালে কোন agglutination হবে না অর্থাৎ negative result পাওয়া যাবে।
>> Control ব্যবহার করে positive হলে কেমন agglutination হবে এবং negative হলে কেমন দেখা যাবে তা বুঝা যায়। তাছাড়া positive এবং negative control ব্যবহার করে reagent এর কার্যকারিতা ঠিক আছে কি না তাও জানা যায়।

Rapid immunochromatographic assay for the qualitative detection of hepatitis B virus antigen অর্থাৎ দেহে Hepatitis B virus এর উপস্থিতি নির্ণয়ের জন্য HBsAg test করা হয়।
Specimen: Serum or plasma.
Test procedure(পরীক্ষা পদ্ধতি):
একটি পরিস্কার test tube এ 300µl থেকে 500µl serum নিতে হবে।
HBsAg strip এর তীর (arrow) চিহ্নিত প্রান্তটি নীচের দিকে রেখে strip টিকে test tube এ রাখা serum এ ডুবাতে হবে।
Strip টিকে এ অবস্থায় ৫মিনিট রাখতে হবে।
Result ( ফলাফল):
HBsAg positive: Strip এর মাঝখানে control band + test band (two bands) দেখা গেলে result positive.
HBsAg negative: Strip এর মাঝখানের উপরের অংশে শুধু control band দেখা গেলে result negative.
Invalid result: নির্দিষ্ট সময় পর যদি strip এ control band না আসে তাহলে result invalid. তখন test টি পুনরায় নতুন strip দিয়ে করতে হবে।
